Bis Tris propane is a zwitterionic buffer primarily used in biochemistry and molecular biology. Because it has two similar pKa values, with a primary acid dissociation constant Pka1=6.8 and a secondary acid dissociation constant Pka2=9.0, it exhibits a very wide buffering range, approximately between pH 6.3 and 9.5. It is precisely because of its broad buffering capacity that the pH can be as low as 6-7, making it more suitable than Tris buffer for improving the stability and activity of restriction endonucleases. It is also one of the reagents with a wide range of applications in the TRIS series of products.
