ApplicationThis product can be used in the water-soluble reagent for the determination of hydrogen peroxid e by enzyme photometry, widely used in diagnosis and biochemical test.

The full name of the enzyme-linked immunosubstrate MADB is N,N-bis(4-sulfobutyl)-3,5-dimethylaniline sodium salt, a peroxidase chromogenic substrate with good hydrophilic properties. Its chromogenic product with 4-AAP has a maximum absorption wavelength of 630 nm and is suitable for the detection of samples with complex composition, e.g. for the enzymatic photometric triglyceride assay.

MADB is an ingredient in biochemical kits and is used as a chromogenic substrate for peroxidase. In the enzymatic photometric triglyceride kit, it is oxidised in the presence of peroxidase, which is coupled to 4-AAP. Triglycerides undergo a series of reactions to produce hydrogen peroxid e oxidation, which can produce a colour development reaction with MADB.

Triglyceride assay principle:
During the assay, triglycerides are first hydrolysed to glycerol and fatty acids in the presence of lipoprotein esterase (LPL, lipoprotein lipase); glycerol and adenosine triphosphate ATP are catalysed by glycerol kinase (GK) to produce glycerol-3-phosphate and ADP; glycerol-3-phosphate and oxygen are catalysed by glycerol-3-phosphate oxidase (GPO) to produce dihydroxyacetone phosphate and hydrogen peroxid e; the hydrogen peroxid e generated is The hydrogen peroxid e generated can then be coupled to the chromogenic substrates MADB and 4-AAP to produce a quinonimine substance, which can then be detected by spectrophotometry at the maximum absorption wavelength of 630 nm, usually by the colourimetric endpoint method.

Triglycerides are esters formed from glycerol and 3 fatty acids and are the most abundant of the natural fats. In the blood, triglycerides bind to apolipoproteins to form very low density lipoproteins (VLDL) and celiac particles, which are transported throughout the body. Triglyceride testing is used to screen for fat status, to detect atherogenic risk and to monitor the effectiveness of lipid-lowering therapy. Recent studies have shown that an increase in triglycerides in combination with an increase in low-density lipoprotein (LDL) is associated with a high risk of coronary heart disease (CHD) in particular. Epidemiological studies have observed that the presence of serum triglycerides >2 mmol/L and high-density lipoprotein cholesterol (HDL-C) <1.0 mmol/L together indicates a high risk of future coronary heart disease (CHD). The clinical application of the triglyceride threshold (>2.3mmol/L) needs to be combined with other risk factors for CHD.
