Tris is called trishydroxymethylaminome thane, also called tromethamine, with CAS number 77-86-1, and its appearance is white crystalline powder. It is soluble in ethanol and water. It also dissolves with ethyl acetate but is relatively slow. It does not react with ether or carbon tetrachloride. It is corrosive to copper and aluminum.
Tris is widely used in acute metabolic and respiratory acidemia. It is an alkaline buffering agent. It has a good buffering effect on metabolic acidosis and enzyme activity. It may cause hypoglycemia, hypotension, nausea, and vomiting. It can even inhibit breathing and stop breathing. Excessive intake or renal insufficiency can cause alkalemia. When Tris and acid titration standard materials are used, the purity requirements will be very high, basically 99.9%-100%
Because Tris is weakly alkaline, when the temperature is below 25ºC, Pka is 8.1, but according to the buffer theory, its effective buffer range is between PH7.0-9.2. The pH of the aqueous solution of Tris is about 10.5, and hydrochloric acid is generally used to adjust the pH to the desired value, but at the same time, attention should be paid to the effect of temperature on the Pka of Tris. Since the Tris buffer is a weakly alkaline solution, DNA will be deprotonated in the solution, thereby improving its solubility. Therefore, people often add EDTA to Tris hydrochloric acid buffer to make TE buffer, and change the pH adjustment solution to acetic acid to obtain TAE buffer.
