HDAOS is a new type of Trinder's reagent with CAS number 82692-88-4, also known as N - (2-hydroxy-3-sulfopropyl) -3'5-dimethoxyaniline sodium salt. HDAOS can be coupled with 4-AAP and oxidized by hydrogen peroxide in the presence of peroxidase, which is a very sensitive reaction. Application:This product can be used in the water-soluble reagent for the determination of hydrogen peroxid e by enzyme photometry, widely used in diagnosis and biochemical test.
Total cholesterol (TC) includes free cholesterol (FC) and cholesterol ester (CE) and can be determined by more than a hundred different methods, which can be divided into five main categories: chemical reagent colourimetric methods, enzymatic methods, fluorescence methods, gas phase and high performance liquid chromatography. Due to the high conditions of gas phase and high performance liquid chromatography, enzymatic determination of total cholesterol using chromogenic substrates such as HDAOS is currently the most routinely applied method, with the advantages of rapidity, accuracy, specificity, low sample volume, no need for extraction and ease of automated analysis and batch determination.
Cholesterol is also known as cholesterol. It is a derivative of cyclopentan e polyhydrophenanthrene. Cholesteryl esters in serum are hydrolysed by the enzyme cholesteryl esterase CHE to free cholesterol and free fatty acids (FFA). Cholesterol is oxidised by the enzyme cholesteryl oxidase CHO to produce 4- cholestenone and hydrogen peroxid e. In the presence of peroxidase, the generated hydrogen peroxid e oxidises the chromogenic substrate HDAOS coupled to 4-AAP to produce a red quinonimine, the absorbance of which is measured by spectrophotometer and is proportional to the TC content of the subject.
The reagents required for the assay of total cholesterol are: pH buffer 50 mmol/L at pH 6.7, cholesterol esterase ≥ 200 U/L, cholesterol oxidase ≥ 100 U/L, peroxidase ≥ 3000 U/L, 4-AAP 0.3 mmol/L, HDAOS 5 mmol/L and also a cholesterol standard solution of approximately 5 mmoL/L.
