Method for Quantitative Determination of Hydrogen Peroxide Using DA-67

1. Prepare 4.08 mg of DA-67 powder, 1 mL100 units/mL of peroxidase, 0.1mol/L, pH=7, Triton X-100 0.5% PIPES buffer;

2. Place the prepared DA-67 powder and peroxidase into PIPES buffer and bring to a constant volume of 100 mL (DA67 100 μ Mol/L solution);

3. Take 3ml of the prepared solution and mix it with 10 μ Mix the sample solution L (H2O2: 0-5 mmol/L);

4. Incubate at 37 ºC for 5 minutes;

5. Measure the absorbance at 666 nm using a spectrophotometer, and use the absorbance difference of the blank (solution mixed with distilled water to replace the sample solution) as the detection value. Use a separately prepared calibration curve to determine the content in the sample.

The Application Principle of DA-67

When used in combination with peroxidase, DA-67 exhibits high sensitivity to hydrogen peroxide and is a water-soluble oxidative chromogenic reagent. Compared to traditional color reagents, it not only has the characteristics of low absorbance and high sensitivity, but also has better water solubility compared to other color reagents DA-67. At a concentration of more than 1 mmol/L, 10 milligrams can be dissolved in 25 milliliters of water. It is compatible with the above advantages and requires a simple colorimetric method to obtain results quickly, greatly increasing detection efficiency.
